Recommended Dilution Suitable for Immunohistochemistry on frozen sections: 7.5 - 30 µg/ml (1/50-1/200).
Suggested positive control: Human tonsil
Antigen Distribution on Isolated cells and Tissue sections: Weakly positive on granulocytes. Positive on peritoneal macrophages. Negative on lymphocytes.
Cell Lines: HL-60: positive on approx. 50% following 1.25% DMSO treatment increased to 100%.
In lymph nodes X-5 is positive on monocytes, macrophages and single cells both intrafollicularly and in surrounding blood vessels. Positive staining has been observed with the basal membrane of blood vessels and on sinus walls. Marginal zone macrophages, medulla and red pulp stain positive in the spleen as do cells in the T-cell region. In the thymus, medulla and trabecular macrophages are positive.

Gene Name integrin subunit alpha M
Background Antibody X-5 recognizes a non-covalently bound alpha heterodimeric glycoprotein. It belongs to the ß-2 integrin supergene family of adhesion. The antigen is a receptor for C3bi, fibrinogen, coagulation factor X. Together with CD18, CD11b forms the MAC-1 molecular complex involved in the adhesion to endothelial cells, and the subsequent extravasation of neutrophils and monocytes.
