Background | DCIR (dendritic cell immunoreceptor), also known as LLIR, DDB27, CLECSF6, HDCGC13P, is a member of the Dectin-2 family of C-type lectins. DCIR is expressed as a type II membrane glycoprotein of 237 aa with a single carbohydrate recognition domain (CRD), closest in homology to those of the macrophage lectin and hepatic asialoglycoprotein receptors. In contrast to the other members of this family, the intracellular domain of DCIR contains a consensus immunoreceptor tyrosine-based inhibitory motif (ITIM). DCIR is expressed on dendritic cells, monocytes, macrophages, B lymphocytes, neutrophils, granulocytes and plasmacytoid dendritic cells, but not detected on NK and T cells. In vitro, DCIR is strongly expressed by DCs derived from blood monocytes cultured with GM-CSF and IL-4 with a higher expression in CD14+ than CD1a+ derived DC. Finally, DCIR expression is down-regulated by signals inducing DC maturation such as CD40 ligand, LPS, or TNF-. Thus, DCIR is differentially expressed on DCs depending on their origin and stage of maturation/activation. DCIR represents a novel surface molecule expressed by antigenpresenting cells, and of potential importance in regulation of DC function. (Bates EE et al, 1999; J. Immunol., 163: 1973-1983). |
