Background | Rotaviruses are RNA viruses from the family Reoviridae that are the the most common cause of gastroenteritis in infants. They have a genome consisting of 11 double stranded RNA segments that code for 6 structural and 5 non-structural proteins. The nucleic acid is contained in a triple layered protein capsid consisting of outer capsid proteins, VP4 and VP7, inner capsid protein -VP6 and core protein -VP2. The five serotypes were purified by high speed centrifugation. The genomic RNA of each preparation was assessed using PAGE, and its group, sub group and serotype assessed by ELISAs, based on a total of eleven different monoclonal antibodies. Thus each serotype was confirmed before immunization. This was done with live Rotavirus serotypes to avoid the risk of surface protein damage (VP7 and VP4). |
