17β-Hydroxysteroid dehydrogenase (HSD17B)
17?-Hydroxysteroid dehydrogenase (HSD17B) are multifunctional enzymes that not only catalyzes the oxidation/reduction of the 17?-hydroxyl/keto group in position C17, C18 and C19 steroids but also execute reactions in intermediary metabolism. 14 isoforms of HSD17B is known. The tissue expression characteristics varies from being tissue specific to ubiquitous. Deficiency in HSD17B4 and HSD17B10 is associated with inborn metabolism errors in humans, which underlines the significance of HSD17B?s role as a metabolic enzyme.

HSD17B2 catalyzes the interconversion of testosterone and androstenedione, as well as estradiol and estrone; in addition to having 20-alpha-HSD activity. It uses NADH as its substrate unlike HSD17B3 which uses NADPH. Studies show that it might play a role in the pathology of breast cancer, prostate cancer, endometriosis and osteoporosis.

HSD17B4 is a bifunctional enzyme that is involved in the peroxisomal beta-oxidation pathway for fatty acids and as a catalyst in the formation of 3-ketoacyl-CoA. Defects in this gene that affect the peroxisomal fatty acid beta-oxidation activity are a cause of D-bifunctional protein deficiency (DBPD). In addition defects in this enzyme is associated with Perrault syndrome and prostate cancer.

HSD17B10 is a mitochondrial protein that catalyzes the oxidation of a wide variety of fatty acids and steroids, and is a subunit of mitochondrial ribonuclease P, which is involved in tRNA maturation. The protein has been implicated in the development of Alzheimer disease, and mutations in the gene are the cause of 17beta-hydroxysteroid dehydrogenase type 10 (HSD10) deficiency.
