Recommended Dilution Suitable for use in precipitating techniques as Immunoelectrophoresis and single and Double Radial Immunodiffusion (Mancini, Ouchterlony) to identify the presence of C1 Esterase Inhibitoror in Human plasma or other body fluids or to determine its concentration. The presence of non-precipitating antibodies has not been assayed. This does not exclude the use of the antiserum in non-precipitating antibody-binding techniques if proper controls are included.
Recommended Dilutions:
Immunoelectrophoresis: Use 2 µl human plasma or equivalent against 120 µl antiserum.
Double Radial Immunodiffusion: Use a rosette arrangement with 10 µl antiserum in 3 mm diameter center well and 2 µl plasma samples (neat and serially diluted) in 2 mm diameter peripheral wells.
Antibody Titre: Precipitin titre 1/32 when tested against normal Human Serum in agar-block immunodiffusion titration.

Specificity The reactivity of the antiserum is restricted to C1 esterase inhibitor.
In Immunoelectrophoresis and Radial Immunodiffusion (Ouchterlony), using various antiserum concentrations against fresh normal Human plasma a single precipitin line is obtained which shows a reaction of identity with the precipitin line obtained with purified C1 esterase inhibitor. No reaction is obtained with any other plasma protein component or serum. However, the antiserum may also react with fibrin monomers, circulating fibrinopeptides and fibrin degradation products.
The antiserum does not cross react with any other component of human plasma. Inter-species cross-reactivity is a normal feature of antibodies to plasma proteins since they frequently share antigenic determinants. Cross-reactivity of this antiserum has not been tested in detail.

Background C1 esterase inhibitor is a single chain acid glycoprotein with a molecular weight of 110,000. The protein has a high carbohydrate content. In immunoelectrophoresis it ha an alpha mobility. The concentration in normal serum ranges from 150 to 350 mg/ml. Reduced levels are observed in hereditary angioedema. It regulates the activation of complement factor C1 as well as the activity of activated C1 by coupling with the active catalytic site at the light chains of C1r and C1s. Moreover it also inhibits, kallikrein, Factor XI and Factor XII.

Note Adsorption: Immunoaffinity adsorbed using insolubilized antigens as required, to eliminate antibodies reacting with other dog serum proteins. The use of insolubilized adsorption antigens prevents the presence of excess adsorbent protein or immune complexes in the antiserum.
