Background | ARK2, an Aurora/IPL1-like protein kinase, functions in bipolar chromosome orientation, kinetochore assembly, spindle checkpoint and microtubule dynamics, and chromosome condensation and cohesion. ARK2 has been shown to be physically associated with inner centromere protein (INCENP) during mitosis, and activated ARK2 has been shown to phosphorylate histone H3. The Ark2 gene has been mapped to 17p13.1, a region that is frequently deleted in tumors, and has been shown to be transcriptionally overexpressed in many tumor cell lines. Overexpression of ARK2 disrupts cleavage furrow formation without affecting nuclear division, often resulting in multinuclearity and polyploidy. |
