Western blot: Approx 16kDa band observed in Human Brain (Frontal Cortex) lysate (calculated MW of 7.6kDa according to NP_006167.1). The observed molecular weights correspond to earlier findings in literature with different antibodies (Li et al, J Biol Chem. 1999 Jan 15;274(3):1294-300; PMID: 9880498). 
Preliminary testing was unsuccessful on Mouse Brain for this particular batch.

Recommended concentration: 0.1-0.3 µg/ml.
Immunohistochemistry on Paraffin Sections: In paraffin embedded Human Brain (Cortex) shows cytoplasm staining in the neuronal cell bodies.

Background Neurogranin (NRGN) is the human homolog of the neuron-specific rat RC3/neurogranin gene. This gene encodes a postsynaptic protein kinase substrate that binds calmodulin in the absence of calcium. The NRGN gene contains four exons and three introns. The exons 1 and 2 encode the protein and exons 3 and 4 contain untranslated sequences. It is suggested that the NRGN is a direct target for thyroid hormone in human brain, and that control of expression of this gene could underlay many of the consequences of hypothyroidism on mental states during development as well as in adult subjects.
