Applications ELISA, WB
Recommended Dilution Direct ELISA: To detect Human IL-33 by Direct ELISA (using 100 μl/well antibody solution) a concentration of ~1.0 μg/ml of this antibody is required. This biotinylated polyclonal antibody allows the detection of at least 0.2–0.4 ng/well of recombinant Human IL-33.
Sandwich ELISA:
To detect Human IL-33 by Sandwich ELISA (using 100 μl/well antibody solution) a concentration of 0.25-1.0 μg/ml of this antibody is required. This Biotin conjuagted antibody, in conjunction with  Anti-Human IL-33 antibody (Cat.-No AP32086PU-S or AP32086PU-N) as a Capture antibody, allows the detection of at least 0.2-0.4 ng/well of recombinant Human IL-33.
Western Blot: To detect Human IL33 use this antibody at a concentration of 0.1-0.2 μg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant Human IL-33 is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.

Background Interleukin 33 (IL-33) is a 32 kDa proinflammatory cytokine and intracellular nuclear factor with transcriptional regulatory properties. IL-33 is structurally related to IL-1, which induces helper T-cells to produce type 2 cytokines and acts through the receptor IL1RL-1 (IL1 receptor-like-1), which is known also as ST2. Binding of IL-33 to this receptor activates NF-kappa-B and MAP kinases and induces in vitro Th2 cells to produce cytokines. In vivo, IL-33 induces expression of IL-4, IL-5, IL-13 and leads to severe pathological changes in mucosal organs and in vitro, it can be divided to N-terminal fragment of 12 kDa and C-terminal fragment of 18kDa by cleavage of caspase-1.
Synonyms IL-33, IL1F11, IL-1F11, NFHEV, NF-HEV, C9orf26
