Applications | ELISA, FN, WB |
Recommended Dilution | ELISA. Indirect: To detect hNP-1 by indirect ELISA (using 100 µl/well antibody solution) a concentration of 0.5-2.0 µg/ml of this antibody is required. This antibody, in conjunction with compatible secondary reagents, allows the detection of at least 0.2-0.4 ng/well of recombinant hNP-1. Sandwich: To detect hNP-1 by sandwich ELISA (using 100 µl/well antibody solution) a concentration of 0.5-2.0 µg/ml of this antibody is required. This antibody, in conjunction with a biotinylated Anti-Human NP-1 (PP1112B) as a detection antibody, allows the detection of at least 0.2-0.4 ng/well of recombinant hNP-1. Western Blot: To detect hNP-1 by Western Blot analysis this antibody can be used at a concentration of 0.1- 0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hNP-1 is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions. Neutralization: To yield one-half maximal inhibition (ND50) of the biological activity of hNP-1 (100 ng/ml), a concentration of 0.40-0.60 µg/ml of this antibody has been used with previous lots. |

Background | Defensins are a group of cyclic peptides containing 29-35 amino acids, which comprise approximately 30% of the total protein content of neutrophil azurophilic granules. Various functions have been reported for the defensins, including antibacterial and antifungal activity and chemotactic activity for monocytes. |
