Efficient Genome Editing with Cas9 mRNA and verified gRNA. HEK293T/17, U2OS and NHDF cells were co-transfected with 0.5 µg of Cas9 encoding mRNA, and 25nM verified gRNA using Turbo-mRNA Transfection Kit (0.5 µl/well of 24-well plate of both mRNA Reagent and Boost, OriGene Catalog TT100314). A T7E1 mismatch detection assay was used to measure cleavage efficiency at 48 hours post-transfection.

  Efficient Genome Editing with Cas9 mRNA and verified gRNA. A defective RFP gene (with deletions) was inserted in the genome of HEK293T cells using CRISPR knockin technology. The RFP negative parent cells were co-transfected with Cas9 encoding mRNA, RFP repairing oligos, and different gRNAs using Turbo-mRNA Transfection Kit (OriGene Catalog TT100314). RFP gene can be repaired by the Cas9 mRNA, verified RFP gRNA and wild type RFP oligos but not scrambled control. RFP positive cells can be detected at 3 days post-transfection. This kit is compatible with serum; transfection efficiency is optimal when transfections are performed in the presence of serum, with no medium change required.
• Storage: Store both Turbo-mRNA Reagent and mRNA Boost Reagent at 4°C. Before each use, warm to room temperature and vortex gently.
• Stability: The transfection reagent is stable for one (1) year from date of shipping when stored at +4°C.
