Immunogen A Synthetic peptide corresponding to amino acids 154-167 of Human MDM2 protein.
Spleen cells from immunised BALB/c mice were fused with cells of the mouse X63Ag8.653 myeloma cell line.
Specificity This antibody recognizes the 90kD MDM2 protein, which forms a complex with p53 leading to p53 inhibition. May also acts to stimulate cell proliferation via its induction of transcription factors such as E2F1 and DPI.
MDM2 is over expressed in a range of human malignancies, including soft tissue sarcomas, breast cancer and bladder cancer.

Background Human homolog of Murine Double Minute-2 (MDM2) and p53 function in mutual regulation: activation of p53 results in expression of MDM2 which consequently inhibits the ability of wild type p53 to stimulate expression of target genes. MDM2 is frequently overexpressed in the nuclei of soft tissue sarcomas, and to a much lesser extent, in a wide variety of malignancies of bladder, breast, testes, cervix, and glial cells.
