Clone Name | 53-6.7 |
Applications | FC, IHC, IP |
Recommended Dilution | Immunoprecipitation. Immunofluorescence. Flow Cytometry: Use 10 µl of the suggested working dilution to label 1x10e6 cells in 100 µl. The Fc region of monoclonal antibodies may bind non-specifically to cells expressing low affinity Fc receptors. Immunohistochemistry on Frozen Sections. Immunohistochemistry on Paraffin Sections: This antibody has been reported as working well on Zinc-fixed paraffin-embedded sections. However, inconsistent results have been reported on formalin-fixed paraffin-embedded tissue sections. |
Reactivities | Mouse |
Host | Rat |
Isotype | IgG2a |
Clonality | Monoclonal |
Immunogen | Mouse thymus or spleen. |
Specificity | This antibody detects CD8a, the alpha chain of the CD8 antigen. CD8 is expressed on T-cells, and exists either a homodimer composed of two alpha chains, or a heterodimer composed of one alpha and one beta chain. The 53-6.7 antibody has been reported to block antigen presentation via MHC class I and inhibit T cell responses to IL-2. It has also been used for Depletion of CD8a+ cells. |
Formulation | PBS, pH 7.2 containing 0.09% Sodium Azide as preservative. State: Purified State: Liquid purified IgG fraction. |
Concentration | 0.5 mg/ml |
Purification | Affinity Chromatography on Protein G from tissue culture supernatant. |

Background | The CD8 antigen is a cell surface glycoprotein found on most cytotoxic T lymphocytes that mediates efficient cell to cell interactions within the immune system. The CD8 antigen, acting as a coreceptor, and the T cell receptor on the T lymphocyte recognize antigen displayed by an antigen presenting cell (APC) in the context of class I MHC molecules. The functional coreceptor is either a homodimer composed of two alpha chains, or a heterodimer composed of one alpha and one beta chain. Both alpha and beta chains share significant homology to immunoglobulin variable light chains. CD8 is expressed on most thymocytes and approximately 1/3 of peripheral blood T cells in humans. CD8 alpha/beta heterodimers are expressed only on TCR alpha/beta T cells, whereas CD8 alpha homodimers can be expressed on alpha/beta and gamma/delta T cells and some NK cells in humans. The pattern of expression in other species has not been well documented. The CD8 alpha chain specifically binds to class I MHC molecules. |
