Background | Nicotinamide mononucleotide adenylyltransferase 1 is an enzyme that in humans is encoded by the NMNAT1 gene. It is an essential enzyme because it catalyzes the final step of NAD+ biosynthesis. There are three isoforms in humans that exhibit tissue- and organelle-specific expression. NMNAT-1 is localized exclusively in the nucleus, whereas NMNAT-2 and NMNAT-3 are found in the Golgi and mitochondria, respectively. NMNAT1 constitute a nuclear NAD+ salvage pathway which regulates the functions of NAD+-dependent enzymes such as the protein deacetylase SIRT1. NMNAT1 protects neurons from neuronal activity induced degeneration. |
