Recommended Dilution | Suitable for Immunohistochemistry and Immunocytochemistry (Frozen or Formalin-Fixed Paraffin-Embedded (FFPE) tissue sections and cell smears) For IHC dilute concentared antibody at 1/50-1/100, use streptavidin~biotin system or polymer system, incubate 30 minutes at room temperature. FFPE tissue section requires antigen retriever (boiling tissue in 10 mM citrate, pH 6.0 for 20 mins, followed by cooling at RT for 20 mins). Immunofluorescence: 10-20 µg/ml (1/10-1/20), incubate for 2 hours in the dark at RT or it can also be incubated overnight at 4ºC. Flow Cytometry: 0.2-1.0 µg/0.1 ml (1/200-1/1,000). Recommended Positive Control: Human colon Cancer. |

Background | P53, also known as protein 53 (TP53), is a transcription factor that regulates the cell cycle and hence functions as a tumor suppressor in a variety of tissue types and is involved in regulating cell growth, replication and apoptosis. It binds mdm2, SV40 T antigen and human papilloma virus E6 protein, p53 senses DNA damage and facilitate response. The human gene that encodes for p53 is TP53. The gene is named TP53 after the protein it codes for (TP53 is another name for p53). The gene is located on the human chromosome 17 (17p13.1. Mutation involving p53 is found in a variety of malignant tumors, including breast, ovarian, bladder, colon, lung and melanoma. |
