• Background: Interleukin 12 (IL-12) is a heterodimeric 70 kD (p70) cytokine comprising two covalently linked glycosylated chains, IL-12a 35 kD (p35) and IL-12b 40 kD (p40). IL-12 is produced by monocytes, macrophages and dendritic cells in response to infection or on interaction with activated T cells. IL-12 induces IFN-γ production, proliferation and cytotoxity in activated natural killer cells and T cells. IL-12 plays a key role in the development of Th1 responses leading to IFN-γ and TNF production, which in turn promote T cell responses and macrophage activation. The IL-12a (p35) chain is shared by IL-35, which is a heterodimer of the IL-12a and IL-27b chains. IL-35 is secreted by regulatory T cells and, in contrast to IL-12, suppresses the inflammatory responses of immune cells.
