Recommended Dilution | Direct ELISA: To detect Human sCD40L by Direct ELISA (using 100 μl/well antibody solution) a concentration of ~1.0 μg/ml is required. In conjunction with compatible secondary reagents, it allows the detection of at least 0.2-0.4 ng/well of recombinant Human sCD40L. Sandwich ELISA: To detect Human sCD40L by Sandwich ELISA (using 100 μl/well antibody solution) a concentration of 0.25-1.0 μg/ml is required. In conjunction with Purified Anti-Human sCD40L (Cat.-No AP01120PU-N or Cat.-No AP01120PU-S) as a Capture antibody, it allows the detection of at least 0.2-0.4 ng/well of recombinant Human sCD40L. Western Blot: To detect Human sCD40L by Western Blot analysis this antibody can be used at a concentration of 0.1-1.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant Human sCD40L is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions. |

Background | CD40 ligand (CD40L) is a 33 kDa type II membrane glycoprotein expressed mainly on the cell surface of activated T lymphocytes, but also exists as a soluble form extracellularly. CD40L is the ligand for CD40, a member of the TNF superfamily, which is expressed on the cell surface of B cells, macrophages/monocytes, dendritic cells, vascular endothelial cells, and epithelial cells. CD40L plays an important role in B cell proliferation, antibody class switching, modulation of apoptosis in the germinal center through interaction with B cells expressing CD40, and activation of CD4+ T cells. Mutation within the CD40L gene is linked to hyper IgM syndrome, an X linked immunodeficiency disease that is characterized by elevated level of serum IgM and decreased level of other isotypes. |
