Recommended Dilution | Peptide ELISA: 1/64000 (Detection Limit). Western blot: Preliminary experiments gave an approx 70kDa band in human lymph node and tonsil lysates after 0.3 µg/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the band we observe given the calculated size of 118kDa according to NP_004886.3 and 106kDa according to NP_899632.1. The 70kDa band was successfully blocked by incubation with the immunizing peptide. Immunohistochemistry on Paraffin Sections: 2-4 µg/ml. In paraffin embedded Human Tonsil shows focal and interstitial staining of the inflammatory elements close to the epithelia of the tonsil crypts. |

Background | NLRP3 is a pyrin-like protein containing a pyrin domain, a nucleotide-binding site (NBS) domain, and a leucine-rich repeat (LRR) motif. This protein interacts with the apoptosis-associated speck-like protein PYCARD/ASC, which contains a caspase recruitment domain, and is a member of the NALP3 inflammasome complex. This complex functions as an upstream activator of NF-kappaB signaling, and it plays a role in the regulation of inflammation, the immune response, and apoptosis. |
