Recommended Dilution Can be used in precipitating techniques as immunoelectrophoresis and single or double radial immunodiffusion (Mancini, Ouchterlony) and electroimmunodiffusion (Laurell) to identify or measure the concentration of MUP. Proteins isolated from urine, such as Bence Jones proteins, may contain MUP as an impurity. This antiserum can be used to identify the impurity and also to remove it by immunoaffinity adsorption. It has not been tested for use in nephelometry, ELISA or immunohistology, but this does not exclude such use if proper controls are included.
Recommedation for Use:
Immunoelectrophoresis: Use 2 ml urine, plasma or equivalent against 120 ml antiserum.
In Double Radial Immunodiffusion (Ouchterlony): Use a rosette arrangement with 10 ml antiserum in a 3 mm diameter centre well and 2 ml serum samples (neat and diluted) in 2 mm diameter peripheral wells.
Precipitin titre: not less than 1/32 when tested against normal Mouse Urine in agarblock immunodiffusion titration.

Immunogen Purified Mouse Major Urinary Proteins are isolated from pooled normal urine.
Freund’s complete adjuvant is used in the first step of the immunization procedure.
Specificity The reactivity of the antiserum is restricted to Mouse MUP. In immunoelectrophoresis and Radial Immunodiffusion, using various antiserum concentrations against normal Mouse plasma characteristic precipitin lines are obtained which show a reaction of identity with the precipitin lines obtained with purified immunogen. No reaction is obtained with any other plasma protein components or serum.
Cross-reactivity: The antiserum does not cross-react with any other component of mouse plasma. Inter-species cross-reactivity is a normal feature of antibodies to plasma proteins since they frequently share antigenic determinants. Cross-reactivity of this antiserum has not been tested in detail.

Reconstitution Method Restore by adding 1 ml of sterile distilled water.
Concentration Total protein and IgG concentrations in the antiserum are comparable to those of pooled normal goat serum. No foreign proteins added.
Background The MUP is a sex related complex with a pre-albumin mobility in gel electrophoresis. Concentrations in male mice are between 50 to 150 mg/ml, in female mice 10 to 20 mg/ml. The complex is also detectable in serum, however concentrations are about 200 to 400 times lower. Variations in the composition of the complex are genetically determined.

Note Adsorption: Immunoaffinity adsorbed using insolubilized antigens as required, to eliminate antibodies crossreacting with other with other proteins. The use of insolubilized adsorption antigens prevents the presence of excess adsorbent protein or immune complexes in the antiserum.
