• Background: Interleukin 10 is a 18kDa protein consisting of 178 residues that is primarily secreted by TH2 clones but also by activated CD4+ and CD8+ T cells, macrophages, natural killer cells and B cells. This anti-inflammatory cytokine both suppresses cytokine production by TH1 clones and inhibits the response of cytokine targeted cells. IL-10 regulates immune mediated inflammation by inhibiting pro-inflammatory cytokine production by macrophages, including TNF-α, IL-1, IL-12 and IFN-γ. Moreover, it can inhibit accessory functions of macrophages in T cell activation which can inhibit T cell-mediated inflammation. IL-10 has also been shown to have a proliferative effect on B cells, thymocytes and mast cells, as well as promoting B cell differentiation into antibody producing cells. 12G8 monoclonal antibody was obtained after rat immunization with human IL10-transfected Cos7 cells.
