Recommended Dilution | Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity of Mouse IL-10 (1.0 ng/ml), a concentration of 0.043-0.064 µg/ml of this antibody is required. Sandwich ELISA: To detect Mouse IL-10 by Sandwich ELISA (using 100 μl/well antibody solution) a concentration of 0.5-2.0 μg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotin conjugated Anti-Murine IL-10 antibody (Cat.-No PP007B1 or PP007B2) as a detection antibody, allows the detection of at least 0.2-0.4 ng/well of recombinant Mouse IL-10. Western Blot: To detect Mouse IL-10 by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant Mouse IL-10 is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions. |

Background | Interleukin 10 (IL-10) is a 178 amino acid protein that is primarily secreted by TH2 clones. IL-10 has dual functions, the first of which is the suppression of cytokine production by TH1 clones responding to antigen presented by monocyte and macrophage antigen presenting cells (APCs). The second function consists of the inhibition of response of cytokine targeted cells, possibly by the downregulation of CD25 (the interleukin-2 receptor) on macrophages and B lymphocytes. IL-10 exerts its biological activity through the IL-10 receptor (IL-10R); a 560 amino acid, 110kDa glycoprotein whose expression can be induced in cultured macrophages and fibroblasts by elevated in HIV-1 infected individuals and has been implicated in the progression of the disease. |
