Specificity MIDC-8 is identifies dendritic leukocytes including epidermal Langerhans cells, veiled cells and interdigitating cells in secondary lymphoid organs, the medulla of the thymus and its in vitro counterparts.
In combination with clone NLDC-145 (cat.no. BM4017) it is very useful for the isolation of dendritic cells from the thymus. Double staining with MIDC-8 and NLDC-145 allows a more precise characterization of cells to be studied. Whereas MIDC-8 recognizes very specific cytoplasmic components, NLDC-145 stains cytoplasm and cell surface.
The MIDC-8 antigen is localized in the cytoplasm as distinct spots near the nucleus. Immunohistological analyses indicate that the antigen recognized by MIDC-8 and NLDC-145 may be biochemically related.
Staining of Isolated Cells:
The antigen is found in dendritic cells. MIDC-8 does not stain thymic epithelial cells which are the major contaminants in thymic dendritic cell preparations. The antigen is also absent in blood cells, bone marrow cells or macrophages isolated from the peritoneal cavity.
Staining of Tissues:
The antigen is found in interdigitating cells in T cell dependent areas of secondary lymphoid organs, the medulla of thymus, veiled cells, and very weakly in Langerhans cells.
Formulation PBS, pH 7.2 containing 10 mg/ml BSA as stabilizer and 0.01% Thimerosal as a preservative.
