Background MD2 is a secreted glycoprotein associating with the extracellular domain of Toll-like receptor 4 (TLR4) and trafficking to and from the cell surface and the Golgi apparatus. It has a signal peptide, two N terminal glycosylation sites, and seven Cys residues and consists of a heterogeneous collection of disulfide-linked oligomers. MD2 is an essential accessory glycoprotein required for LPS recognition and associates with toll-like receptor 4 on the cell surface to confer responsiveness to lipopolysaccyaride (LPS), and hence provides a link between the receptor and LPS signaling. It can also function as a soluble receptor for cells that do not otherwise express it. MD2 contains multiple trypsin cleavage sites and its expression is correlated inversely with trypsin activity.
