Background | As the only member of intercrine delta/ CX3C family, CX3CR1 (C-X3-C motif chemokine 1) is a type I transmembrane protein with CX3C chemokine domain present on an extended glycosylated stalk and it share this unique membrane anchorage with CXCR6 ligand, CXCL16. CX3CL1 is constitutively shed from the cell surface by ADAM10 (A Disintegrin and Metalloprotease 10) and CX3CL1 cleavage is enhanced upon activation of the closely related protease TNFalpha-converting enzyme, TACE (ADAM-17). CX3CL1 is expressed in a wide variety of cell types including neurons, intestinal , airway , skin epithelium cells, airway smooth muscle cells, and endothelium. It is induced by TNF and IL1 in pulmonary endothelial cells and umbilical vein endothelial cells. CX3CL1 signals through CX3CR1 (the only receptor of CX3CL1) which is expressed on monocytes, natural killer cells, T cells and smooth muscle cells. The major function of CX3CL1's soluble form is to act as chemotactic for T-cells and monocytes, but not for neutrophils, whereas, the membrane-bound form promotes adhesion of those leukocytes to endothelial cells where it may play a role in regulating leukocyte adhesion and migration processes at the endothelium. In addition to its role in chemotaxis and adhesion of leukocytes, CX3CL1 implicates in survival of multiple cell types during homeostasis and inflammation. |
