Background | CARD12, also known as CLAN, IPAF and CLAN1 is a member of the CED4 family of proteins. The CED4/Apaf-1 proteins are critical regulators of apoptosis and NF-kappaB signaling pathways, and CARD12 is no exception. CARD12 has been implicated in coordination of downstream activation of both apoptotic and inflammatory signaling pathways (PMID: 11374873). Additionally, CARD12 has shown to coprecipitate with caspase 1, an enzyme that participates in both the signaling of apoptosis and cytokine processing, further demonstrating its integral role in programmed cell death (PMID: 11374873). As part of the nucleotide-binding site and leucine-rich repeat (NBS-LRR) proteins, CARD12 is known to be an important regulator of caspase-1 activation via an ATP/dATP nucleotide-binding pocket (PMID: 15882992). CARD12 has four known isoforms. Isoform 1 is highly expressed in lung, followed by leukocytes (especially monocytes), lymph node, colon, brain, prostate, placenta, spleen, bone marrow and fetal liver. Although it is expressed ubiquitously, Isoform 2 is more prominent at the highest levels in lung and spleen. Isoform 4 can only been found in the brain. |
