Gene Name NLR family, pyrin domain containing 3
Background NALP3 (NACHT, LRR and PYD domains-containing protein 3, or NLRP3) belongs to the NLRP family and it contains one DAPIN domain, seven LRR (leucine-rich) repeats and one NATCH domain. It interacts with the apoptosis-associated speck-like protein PYCARD/ASC, which contains a caspase recruitment domain and is a member of the NALP3 inflammasome complex. The NALP3 complex functions as an upstream activator of NF-kappaB signaling and it plays a role in the regulation of inflammation, the immune response and apoptosis. It is also reported that the NALP3 inflammasome can be activated by pathogen-associated molecular patterns (PAMPs) or endogenous danger-associated molecular patterns (DAMPs). The activation of the NALP3 inflammasome results in proteolytic activation and secretion of cytokines of the interleukin-1 (IL-1) family. Mutations in this gene are associated with familial cold autoinflammatory syndrome (FCAS), Muckle-Wells syndrome (MWS), chronic infantile neurological cutaneous and articular (CINCA) syndrome and neonatal-onset multi-system inflammatory disease (NOMID).
