• Background: LAMP2 (Lysosome-associate protein 2) is one of a family of membrane glycoproteins whose function is to provide selectins with carbohydrate ligands. LAMP2 has been implicated in tumor metastasis, as well as overall protection, maintenance and adhesion of the lysosome. Specifically, LAMP2 may protect the lysosomal membrane from auto digestion, and also maintain the acidic environment necessary for the lysosome to function correctly. On a subcellular level, LAMP2 is expressed as a single-pass type I membrane protein, shuttling between endosomes, lysosomes, and the plasma membrane. Mutations in LAMP2 lead to a lysosomal glycogen storage disease known as "Danon Disease". Recently, caspase activation has been suggested in the extracellular export of autophagic vesicles, such as LAMP2 (PMID: 22692030).
