Background | Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMP's are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. Unpaired specific MMPs by tissue inhibitor of MMPs (TIMPs) are related in the invasive tumor processes. Interstitial collagenase (MMP-1)(52 kDa), gelatinase A/type IV collagenase (MMP-2) (72 kDa), and gelatinase B/type V collagenase (MMP-9) (92 kDa) dissolve extracellular matrix (ECM) and may initiate and promote angiogenesis. Human collagenase-3 (MMP13) is a recently identified member of the matrix metalloproteinase (MMP) family that is expressed in breast carcinomas and in articular cartilage from arthritic patients. This protein is secreted as a 60 kDa proenzyme and active form of 48 kDa by cleavage. |
