Flow-cytometry using anti-CD44 antibody Hermes-3 (Ab00629) Human lymphocytes were stained with an isotype control (Ab00102-23.0, panel A) or the rabbit-chimeric version of Hermes-3 (TA386030, panel B) at a concentration of 1 µg/ml for 30 mins at RT. After washing, bound antibody was detected using a AF488 conjugated donkey anti-rabbit antibody (ab150073) and cells analysed on a FACSCanto flow-cytometer.

  This antibody has been used in IF, IHC (immunoperoxidase method), cross-blocking and WB studies to characterise the Hermes-defined antigens (Picker, 1989). This antibody is able to block lymphocyte binding to mucosal HEV, without affecting binding to HEV in peripheral lymph nodes and synovium (Jalkanen, 1987). H3-51 is cross-blocked by this antibody, indicating that the epitopes recognized by these two antibodies overlap (Picker, 1989).
