Immunogen | BALB/c mice were immunized with human T lymphoblasts and spleen cells of the immunized animals were fused with X63-Ag8.653 mouse myeloma cells. The supernatants of the resultant hybridomas were screened by a proliferation inhibition assay (inhibition of the human T lymphoblast response to IL-2). Two of those (the clones AHT54 and AHT107) that produced antibodies which inhibited IL-2 dependent proliferation of human T lymphoblasts were selected, cloned and recloned. |
Specificity | AHT54 recognises a determinant close to the IL-2 binding site. AHT54 has been used in ELISAs for the detection of soluble human IL-2R in response to different forms of helminthic and protozoal infections (O. Josimovic et al. 1988). AHT54 has also been shown to have anti-proliferative activities, and chimeric AHT54 antibodies with a human contant region have an increased inhibitory effect on proliferation of T lymphocytes (Rose 1992). |
