HEK293T cells were transfected with the pCMV6-ENTRY control (Left lane) or pCMV6-ENTRY STAT3 (RC212394, Right lane) cDNA for 48 hrs and lysed. Equivalent amounts of cell lysates (5 ug per lane) were separated by SDS-PAGE and immunoblotted with anti-STAT3. Positive lysates LY403728 (100ug) and LC403728 (20ug) can be purchased separately from OriGene.

  Equivalent amounts of cell lysates (10 ug per lane) of wild-type Hela cells (WT, Cat# LC810HELA) and STAT3-Knockout Hela cells (KO, Cat# LC810228) were separated by SDS-PAGE and immunoblotted with anti-STAT3 monoclonal antibody TA500177. Then the blotted membrane was stripped and reprobed with anti-b-actin antibody (TA811000) as a loading control (1:500).

• Background: The protein encoded by this gene is a member of the STAT protein family. In response to cytokinesand growth factors, STAT family members are phosphorylated by the receptor associated kinases, andthen form homo- or heterodimers that translocate to the cell nucleus where they act astranscription activators. This protein is activated through phosphorylation in response to variouscytokines and growth factors including IFNs, EGF, IL5, IL6, HGF, LIF and BMP2. This proteinmediates the expression of a variety of genes in response to cell stimuli, and thus plays a keyrole in many cellular processes such as cell growth and apoptosis.
