HEK293T cells were transfected with the pCMV6-ENTRY control (Cat# PS100001, Left lane) or pCMV6-ENTRY SPHK1 (Cat# RC203398, Right lane) cDNA for 48 hrs and lysed. Equivalent amounts of cell lysates (5 ug per lane) were separated by SDS-PAGE and immunoblotted with anti-SPHK1(Cat# TA507215). Positive lysates LY411847 (100ug) and LC411847 (20ug) can be purchased separately from OriGene.

  Western blot analysis of extracts (30ug per lane) from 2 cell lines lysates by using anti-SPHK1 monoclonal antibody (TA507215, 1:2000).

• Background: The protein encoded by this gene catalyzes the phosphorylation of sphingosine to form sphingosine-1-phosphate (S1P), a lipid mediator with both intra- and extracellular functions. Intracellularly, S1P regulates proliferation and survival, and extracellularly, it is a ligand for cell surface G protein-coupled receptors. This protein, and its product S1P, play a key role in TNF-alpha signaling and the NF-kappa-B activation pathway important in inflammatory, antiapoptotic, and immune processes.
