Background | 17β-Hydroxysteroid dehydrogenase type 13 (HSD17B13) is an enzyme that in humans is encoded by the HSD17B13 gene. HSD17B13 is a 300-amino-acid protein that is localized on lipid droplet. The RNA expression of HSD17B13 is highly restricted to the liver, specifically in hepatocytes but not other cell types in the liver. HSD17B13 is involved in various metabolic processes, including steroid hormones, fatty acids, cholesterol, and bile acids. HSD17B13 has been reported to catalyze multiple substrates including steroids, bioactive lipids including leukotriene B4 and 12(R)-hydroxyeicosatetraenoic acid, and retinol. HSD17B13 enzymatic activity attributes to hepatic cell injury, inflammation, and fibrosis, chronically leading to the development of alcoholic liver disease (ALD), nonalcoholic steatohepatitis (NASH), cirrhosis, and even hepatocellular carcinoma (HCC). HSD17B13 expression is significantly upregulated in patients and mice with non-alcoholic fatty liver disease (NAFLD). Hepatic overexpression of HSD17B13 promotes lipid accumulation in the liver. HSD17B13 has potential as a biomarker of advanced liver disease, such as non-alcoholic steatohepatitis (NASH) and liver cancer. |
