HEK293T cells were transfected with the pCMV6-ENTRY control (Left lane) or pCMV6-ENTRY PSMA6 (RC209976, Right lane) cDNA for 48 hrs and lysed. Equivalent amounts of cell lysates (5 ug per lane) were separated by SDS-PAGE and immunoblotted with anti-PSMA6. Positive lysates LY419119 (100ug) and LC419119 (20ug) can be purchased separately from OriGene.

  Western blot analysis of extracts (35ug) from 9 different cell lines by usin g anti-PSMA6 monoclonal antibody (HepG2: human; HeLa: human; SVT2: mouse; A549: human; COS7: monkey; Jurkat: human; MDCK: canine; PC12: rat; MCF7: human).

• 

  HEK293T cells transfected with either RC209976 overexpress plasmid (Red) or empty vector control plasmid (Blue) were immunostained by anti-PSMA6 antibody (TA800104), and then analyzed by flow cytometry.

• Background: The proteasome is a multicatalytic proteinase complex with a highly ordered ring-shaped 20S core structure. The core structure is composed of 4 rings of 28 non-identical subunits; 2 rings are composed of 7 alpha subunits and 2 rings are composed of 7 beta subunits. Proteasomes are distributed throughout eukaryotic cells at a high concentration and cleave peptides in an ATP/ubiquitin-dependent process in a non-lysosomal pathway. An essential function of a modified proteasome, the immunoproteasome, is the processing of class I MHC peptides.
