HEK293T cells were transfected with the pCMV6-ENTRY control (Left lane) or pCMV6-ENTRY TNFRSF4 (RC211253, Right lane) cDNA for 48 hrs and lysed. Equivalent amounts of cell lysates (5 ug per lane) were separated by SDS-PAGE and immunoblotted with anti-TNFRSF4 (1:1000).

  Western blot analysis of extracts (35ug) from 2 cell lines (Human PBMC treated with 10ug/ml PHA for 72h (Right)/untreated (Left)) lysates by using anti-TNFRSF4 monoclonal antibody (1:500).

  Flow cytometric analysis of living 293T cells transfected with TNFRSF4 overexpression plasmid (RC211253), Red)/empty vector (PS100001, Blue) using anti-TNFRSF4 antibody (UM800167). Cells incubated with a non-specific antibody (Green) were used as isotype control. (1;100)

  Flow cytometric analysis of living PBMCs treated with 10ug/ml PHA for 72h (Right)/untreated (Left) using anti-TNFRSF4 antibody (UM800167) (1:100).

• Background: The protein encoded by this gene is a member of the TNF-receptor superfamily. This receptor has been shown to activate NF-kappaB through its interaction with adaptor proteins TRAF2 and TRAF5. Knockout studies in mice suggested that this receptor promotes the expression of apoptosis inhibitors BCL2 and BCL2lL1/BCL2-XL, and thus suppresses apoptosis. The knockout studies also suggested the roles of this receptor in CD4+ T cell response, as well as in T cell-dependent B cell proliferation and differentiation. [provided by RefSeq, Jul 2008]
