Applications ELISA, ID, IF, IHC, IP, WB
Recommended Dilution Can be used in Immunocytochemical and Immunohistochemical staining for the detection of IgD at the cellular and subcellular level by staining of appropriately treated cell and tissue substrates, and to demonstrate circulating antibodies in serodiagnostic microbiology.
In non-isotopic assay methodology (e.g. ELISA) to identify and measure IgD in human serum or other body fluid.
As a second step an avidin or streptavidin conjugate of the user’s choice has to be used.
This immunoconjugate is not pre-diluted. The optimum working dilution of each conjugate should be established by titration before being used. Excess labelled antibody must be avoided because it may cause high unspecific background staining and interfere with the specific signal.
Recommended Dilutions:
Histochemical and Cytochemical Use: 1/100-1/250
ELISA and comparable non-precipitating antibody-binding assays: 1/1,000-1/10,000.
Performance testing. Isotype specificity and quantitative specific recognition ability are further evaluated at the high level of sensitivity by direct singe and simultaneous double staining of different types of human cells, using counterstaining with reference conjugates with a different marker. The immunoconjugate is further tested in ELISA-type assays.
Reactivities Human
Host Goat
Isotype IgG
Clonality Polyclonal
Immunogen Highly purified monoclonal IgD isolated from Human serum. Freund’s complete adjuvant is used in the first step of the immunization procedure.
Specificity The reactivity of the antiserum is directed to the Fc subunits of the IgD molecule. It does not react with any non-Ig protein in human serum, as tested by Immunoelectrophoresis and double radial immunodiffusion.
Cross-reactivity: Inter-species cross-reactivity is a normal feature of antibodies to immunoglobulins, since Ig of different species frequently share antigenic determinants. Cross-reactivity of this antiserum has not been tested in detail.
Formulation PBS, pH 7.2 with no preservative
Label: Biotin
State: Lyophilized hyperimmune purified Ig fraction
Molar radio: Biotin/IgG ~6.0
Reconstitution Method Restore by adding 1.0 ml sterile distilled water
Concentration 10.0 mg/ml
Purification Hyperimmune antisera with strong precipitating activity are selected for fractionation by salt-precipitation and purification of the IgG (7S) fraction by DEAE-chromatography.
Conjugation Biotin
Storage Store lyophilized at 2-8°C and reconstituted at 2-8°C for one week or (in aliquots) at -20°C for longer.
Avoid Repeated thawing and freezing.
Note Adsorption: Immunoaffinity adsorbed using insolubilized antigens as required to eliminate antibody activity to the common Fab portion of immunoglobulins and to any other serum protein. The use of insolubilized adsorption antigens prevents the presence of excess adsorbent protein or immune complexes in the antiserum.
