Recommended Dilution | Can be used in Immunocytochemical and Immunohistochemical staining for the detection of IgE at the cellular and subcellular level by staining of appropriately treated cell and tissue substrates, and to demonstrate circulating antibodies in serodiagnostic microbiology. In non-isotopic assay methodology (e.g. ELISA) to identify and measure IgE in mouse serum or other body fluid. As a second step an avidin or streptavidin conjugate of the user’s choice has to be used. This immunoconjugate is not pre-diluted. Excess labelled antibody must be avoided because it may cause high unspecific background staining and interfere with the specific signal. Recommended Dilutions: Histochemical and Cytochemical Use: 1/100-1/250. ELISA and comparable non-precipitating antibody-binding assays: 1/500-1/2,000. |

Specificity | The reactivity of the antiserum is directed to the Fc subunits of the IgE molecule. It does not react with any non-Ig protein in mouse serum, as tested by Immunoelectrophoresis and double Radial Immunodiffusion. Inter-species cross-reactivity is a normal feature of antibodies to immunoglobulins, since Ig of different species frequently share antigenic determinants. Cross-reactivity of this antiserum has not been tested in detail. Performance Testing: Isotype specificity and quantitative specific recognition ability are further evaluated at the high level of sensitivity by direct singe and simultaneous double staining of different types of Mouse cells, using counterstaining with reference conjugates with a different marker. The immunoconjugate is further tested in ELISA-type assays. |

Note | Adsorption: Immunoaffinity adsorbed using insolubilized antigens as required, to eliminate antibodies cross-reacting with other with other plasma proteins. The use of insolubilized adsorption antigens prevents the presence of excess adsorbent protein or immune complexes in the antiserum. |
