Recommended Dilution Can be used in precipitating techniques as immunoelectrophoresis and radial immunodiffusion to identify the presence of IgG1 in bovine serum and other body fluids or to determine its concentration. To prepare an immunoadsorbent for the purification of bovine IgG1 from serum or plasma.
Antibody titre: Precipitin titre not less then 1/16 when tested against pooled normal bovine serum in agar block immunodiffusion titration.
Recommended Dilutions:
Immunoelectrophoresis: 2 µl or equivalent against 120 µl antiserum.
Double Radial Immunodiffusion (Ouchterlony): use a rosette arrangement with 10 µl antiserum in a 3 mm diameter centre well and 2 µl serum samples (neat and diluted) in 2 mm diameter peripheral wells.
Reactivities Bovine
Host Rabbit
Clonality Polyclonal
Immunogen Pools of purified normal IgG1 isolated from pooled Bovine Serum.
Freund’s complete adjuvant is used in the first step of the immunization procedure.
Specificity The reactivity of the antiserum is directed to the Fc subunit of subclass IgG1. It does not react with other IgG2, IgG/Fab fragments, IgM and IgA or any non-Ig protein in Bovine serum, as tested by immunoelectrophoresis and Double Radial Immunodiffusion.
Cross-reactivity: This IgG fraction is not species-specific since inter-species cross-reactivity is a normal feature of antisera to immunoglobulins. Cross-reactivity of this product has not been tested in detail.
Formulation State: Serum
State: Delipidated, heat inactivated, lyophilized whole antiserum - No preservative added
Reconstitution Method Restore with 1.0 ml distilled water
Concentration Protein concentration Total protein and IgG concentration in the antiserum are comparable to those of pooled normal rabbit serum. No foreign proteins added.
Storage Store lyophilized at 2-8°C for 6 months or at -20°C long term.
After reconstitution store the antibody undiluted at 2-8°C for one month
or (in aliquots) at -20°C long term.
Avoid repeated freezing and thawing.
Note Adsorption: Immunoaffinity adsorbed using insolubilized antigens as required to eliminate antibodies cross-reacting with other components of the immunoglobulin system or reacting with other serum proteins. Special attention is given to the removal of antibodies to common Ig/Fab. The use of insolubilized adsorption antigens prevents the presence of excess adsorbent protein or immune complexes in the antiserum.
