Applications ELISA, ID, IF, IHC, IP, WB
Recommended Dilution Can be used in enzyme-immunocytochemical and immunohistochemical staining for the detection of antigens or antibodies at the cellular and subcellular level by staining of appropriately treated cell and tissue substrates; to demonstrate circulating antibodies in serodiagnostic microbiology and autoimmune diseases; to identify a specific antigen using a reference antibody of monkey origin in the middle layer of the indirect test procedure; in non-isotopic assay methodology (e.g. ELISA, Western blotting). This immunoconjugate is not pre-diluted. The optimum working dilution of each conjugate should be established by titration before being used. Excess labelled antibody must be avoided because it may cause high unspecific background staining and interfere with the specific signal.
Recommended Working Dilutions:
For histochemical and cytochemical use are usually between 1/100 and 1/500.
In ELISA and comparable non-precipitating antibody-binding assays between 1/2000 and 1/10000.
Reactivities Monkey
Host Goat
Isotype IgG
Clonality Polyclonal
Immunogen Purified normal IgG isolated from pooled Rhesus Monkey serum.
Freund’s complete adjuvant is used in the first step of the immunization procedure.
Specificity The reactivity of the antiserum is directed to the Fab and Fc subunits of the IgG. It includes a certain degree of reactivity with other immunoglobulins via the common Fab portion. It does not react with any non-Ig protein in monkey serum, as tested by immunoelectrophoresis and double radial immunodiffusion.
Cross-reactivity: Inter-species cross-reactivity is a normal feature of antibodies to immunoglobulins, since Ig of different species frequently share antigenic determinants. Cross-reactivity has not been tested in detail, but reactions with IgG in serum of other old-world monkeys, including Cercopithecus, Cynomolgus and baboon, chimpanzee and man can be expected.
Formulation PBS, pH 7.2
No preservative added, as it may interfere with the antibody activity. No foreign proteins added.
Label: HRP
State: Lyophilized purified Hyperimmune IgG fraction
Label: Horseradish Peroxidase.
Molar radio: ~1.7
Reconstitution Method Restore with 1 ml sterile distilled water
Concentration 8.4 mg/ml
Purification DEAE-column Chromatography
Conjugation HRP
Storage Prior to reconstitution store at 2-8°C.
Following reconstitution store undiluted at 2-8°C for one week
or (in aliquots) at -20°C for longer.
Avoid repeated freezing and thawing.
Note Enzyme Marker: Horseradish peroxidase enriched for isoenzyme C (RZ=3.2).
Conjugation Procedure: Conjugation is carried out using a proprietary modification of the periodate technique for the binding to peroxidase, followed by several purification steps. After each step activity and specificity are tested in a variety of techniques. The conjugate is lyophilized to assure stability and long shelf life.
