Background FTO (alpha-ketoglutarate-dependent dioxygenase FTO) is a novel protein belonging to FTO family and it is a nuclear dioxygenase that repairs alkylated DNA and RNA by oxidative demethylation reactions. FTO exerts highest demethylase activity towards single-stranded RNA containing 3-methyluracil, followed by single-stranded DNA containing 3-methylthymine, low activity towards single-stranded DNA containing 1-methyladenine or 3-methylcytosine and is inactive towards 1-methylguanine or double-stranded DNA. FTO requires O2, alpha-ketoglutarate and iron (co-factor) for its activity and gets activated by ascorbate whereas its inhibitors include N-oxalylglycine, fumarate, succinate etc. It contributes to the regulation of global metabolic rate, energy expenditure/homeostasis and has been suggested to control the body size as well as accumulation of body fat. FTO has been associated with various obesity traits, insulin resistance and NIDDM, and its defects are known to cause growth retardation developmental delay coarse facies and early death (GDFD).
