HEK293T cells were transfected with the pCMV6-ENTRY control (Left lane) or pCMV6-ENTRY AMACR (RC213437, Right lane) cDNA for 48 hrs and lysed. Equivalent amounts of cell lysates (5 ug per lane) were separated by SDS-PAGE and immunoblotted with anti-AMACR. Positive lysates LY402314 (100ug) and LC402314 (20ug) can be purchased separately from OriGene.

  Equivalent amounts of cell lysates (10 ug per lane) of wild-type Hela cells (WT, Cat# LC810HELA) and AMACR-Knockout Hela cells (KO, Cat# LC810338) were separated by SDS-PAGE and immunoblotted with anti-AMACR monoclonal antibody TA808476. Then the blotted membrane was stripped and reprobed with anti-HSP90AB1 antibody (TA500494) as a loading control (1:500).

• Background: This gene encodes a racemase. The encoded enzyme interconverts pristanoyl-CoA and C27-bile acylCoAs between their (R)- and (S)-stereoisomers. The conversion to the (S)-stereoisomers is necessary for degradation of these substrates by peroxisomal beta-oxidation. Encoded proteins from this locus localize to both mitochondria and peroxisomes. Mutations in this gene may be associated with adult-onset sensorimotor neuropathy, pigmentary retinopathy, and adrenomyeloneuropathy due to defects in bile acid synthesis. Alternatively spliced transcript variants have been described.
