Secretory IgA (sIgA) functions as a dimer or polymer and accounts for almost all specific mucosal antibody activity. A molecule of sIgA is made up of two molecules of IgA, one J chain and one SC (MW 65,000). The dimer IgA is transported into secretions by its binding to the SC on the epithelial cells. Under normal conditions, sIgA contains both subclasses IgA1 and IgA2, since both are capable of binding SC. SC also has an affinity for polymeric IgM. Purified free human secretory component isolated from pooled milk is used for immunization.

Applications ELISA: In Sandwich ELISA a 1/800 dilution of GGHL-30PH-2 was found to generate an O.D of 1.0 in a 20 minute reaction with p-Nitrophenyl Phosphate as the substrate.
Western Blot (1/1000).
Immunoelectrophoresis: Chicken serum (3 µl) was placed in the center well (at the clear circle) and then subjected to electrophoresis. After separation, Goat anti-Chicken serum (75 μl) was placed in the lower trough and GGHL-30 (75 μl) (pre-conjugated affinity-purified antibody) was placed in the upper trough. After overnight incubation at 4°C, the gel was washed, fixed and stained with Coomassie. Note the single precipitin line between the center well and upper trough.
Quality Control: Antibodies were analyzed using Immunoelectrophoresis and a Sandwich ELISA.

Recommended Working Dilutions:
Immunoelectrophoresis: Use 2 µl or equivalent against 120 µl antiserum.
Double Radial Immunodiffusion (Ouchterlony): Use a rosette arrangement with 10 µl in a 3 mm diameter center well and 2 µl serum samples (neat and serially diluted) in 2 mm diameter peripheral wells.
